Nerve Terminal Staining Kits contain Biotium’s nerve terminal dyes and background reducing agents for washing away or quenching extracellular dye fluorescence. Kits are available with SynaptoGreen™ C4 (equivalent to FM®1-43), the fixable green nerve terminal dye AM1-43, or SynaptoRed™ C2 (equivalent to FM®4-64).
The kits are available with a choice of background reducing agents. ADVASEP-7 is a sulfonated cyclodextrin derivative that has been reported to reduce background fluorescence when using SynaptoGreen™ C4 to stain brain slices. SCAS (4-Sulfonato calix[8]arene, sodium salt) is a quencher developed by Biotium to reduce background fluorescence. Unlike ADVASEP-7, SCAS dramatically lowers background fluorescence as soon as it is added to the preparation without the need for repeated wash steps. Sulforhodamine 101 has been used as a quencher dye to reduce SynaptoGreen™ C4 fluorescence.
